| 目的 观察IL-12 siRNA对脑胶质瘤细胞增殖与凋亡的影响.方法将含有IL-12 siRNA的表达质粒稳定转染人脑胶质瘤细胞,应用MTT法及AO/EB染色观察脑胶质瘤细胞的增殖与凋亡情况.结果转染后,MTT法显示IL-12 siRNA组中的细胞生长受抑明显,AO/EB染色显示IL-12siRNA组的凋亡率较阴性对照组及空载体组高.结论 IL-12siRNA靶向干扰了IL-12蛋白的表达,并对脑胶质瘤细胞具有抑制增殖和促进凋亡的作用. |

| Objective To observe the effect of IL - 12siRNA on the proliferation and apoptosis of human gliom~, cell. Methods We transfected human brain glioma cells with the plasmid containing IL - 12siRNA gene. Test the proliferation and apoptosis of glioma cell by using the method of MTr and AO/EB staining . Results The proliferation of gliomacell in intervention group （ IL - 12siRNA） was significantly inhibited after transfection. An we observed more apoptosis in intervention group through AO/EB staining, the apoptosis rate of intervention group significantly higher than that of control group and empty vector group. Conclusions IL - 12siRNA depressed the expression of IL - 12 proteins, meanwhile itcould inhibit proliferation and promote apoptosis of human glioma cell. |
